Studies on Transfer Ribonucleic Acid-Ribosome Complexes XXI. EFFECT OF ANTIBIOTICS ON PEPTIDYL-PURO?(/IYCIN SYNTHESIS BY MAMMALIAN POLYRIBOSOMES*
نویسندگان
چکیده
The effect of antibiotics and other agents on-peptide bond synthesis by mammalian polyribosomes was determined by studying peptidyl-puromycin synthesis. Sparsomycin, anisomycin, cycloheximide, gougerotin, blasticidin S, and cryptopleurine inhibited the reaction. Sparsomycin and anisomytin inhibited the peptidyl-puromycin synthesis competitively with respect to puromycin. Arabinose analogs of puromycin as well as many other antibiotics were ineffective as inhibitors of this reaction. In addition, the results demonstrated that antibiotics and other agents influence peptide bond formation on polyribosomes differently than peptide bond synthesis as measured by acetyl-phenylalanyl-puromycin synthesis. It is thus suggested that the former assay is probably more representative of the mode of action of these agents in the intact cell than the latter.

Antibiotics were used as probes of ribosome topology and function. Studies of [(14)C]chloramphenicol and [(14)C]erythromycin binding to ribosomes and polyribosomes revealed the following features. The requirement of high K(+) concentration (150 mM) for [(14)C]chloramphenicol binding to NH(4)Cl-washed ribosomes resulted from the washing procedure. Neither native 70S ribosomes nor polyribosomes r...

Protein synthesis is the result of a series of coupled events, many of which can be measured. In this report, the determination of the synthesis of peptides of chain length three and greater was used as a measure of translocation, the movement of the ribosome along the polyribonucleotide template. Translocation was observed both in the presence and absence of one added soluble transfer factor w...

Streptomycin-sensitive 70s ribosomes which are dissociated into SOS and 30s particles by dialysis against 1 X lop4 M magnesium acetate, and then reassociated in 0.02 M magnesium acetate, differ from nondissociated 70s ribosomes in r4C-aminoacyl soluble ribonucleic acid (sRNA) binding. 1. Reassociated ribosomes bind about twice as much 14C!phenylalanyl-sRNA in the presence of polyuridylic acid a...
